Improved Physical Performance Parameters in Patients Taking Nicotinamide Mononucleotide (NMN): A Systematic Review of Randomized Control Trials
Published 2024Reviewer Insight3/22/2026Wrong product tested: This study tested NMN (a molecule the body converts to NAD+), not liposomal NAD+ — these are different supplements with different absorption mechanisms
Brain health not studied: The research only measured physical outcomes like walking speed and grip strength — brain health was never assessed
Results were not significant: Even for physical performance, the authors concluded improvements were statistically nonsignificant compared to placebo
Bottom line: This paper cannot support the claim — it tests a different compound, measures different outcomes, and found no significant effects even for what it did measure
- Tested in Humans
Efficacy of oral nicotinamide mononucleotide supplementation on glucose and lipid metabolism for adults: a systematic review with meta-analysis on randomized controlled trials
Published 2024Reviewer Insight3/22/2026NMN is not the same as liposomal NAD+: This study tested a NAD+ building block (NMN) in standard pill form — not the liposomal delivery format claimed, making direct comparison imperfect
Brain health was never measured: Not a single study in this review tested memory, cognition, or any brain outcome — the "brain health" part of the claim has zero human trial evidence here
Blood NAD goes up, but health doesn't improve: NMN does raise NAD in the blood, but this didn't translate to better blood sugar, cholesterol, body weight, or physical performance in 513 people across 12 trials
Most studies had quality problems: 5 of 12 trials had high risk of bias, and there's statistical evidence that negative results were less likely to be published — meaning even these weak results may be optimistic
Bottom line: The best human evidence available shows NMN supplementation raises NAD in blood but does not improve the cellular health markers tested, and brain health specifically has never been tested in humans.
